培养基模拟灌装，未充分挑战人员疲劳，缺陷案例!

近日，FDA给礼来的483缺陷报告中，提及培养基模拟灌装期间没有充分挑战人员疲劳的缺陷：

Adequate justification was not provided tosupport of how the conditions simulated during your Fill Duration Challenge -NLT(b) (4) in Media fill MF0116 - MF0271 , D291263 is reflective of routinemanufacturing.

Fatigue is not adequately challenged.Filling Operator Extended Personnel Shift was listed as being challenged for 14hours, 9 minutes (Protocol Required Challenge NLT XX) during MF0273, BatchD256292 per APS Summary report, effective July 6, 2020. An aseptic operator shift is [ b) (4) i · The media fill D256292 did not support the operator working on the aseptic 1ine for 14 hours, 9 minutes. Management confirmed theoperator did not have to work on the aseptic fill line for the entire time theyare challenging fatigue. They stated they do not consider the time the operatoris working in the aseptic B 103 vial filling line during the fatigue challenge,instead the monitor the length of the operators shift, regardless of where theyare working. As performed by the firm, the fatigue challenge does not ensurethe operator maintains aseptic technique even when they are fatigued.
